Growth and Development of Human Primary Oocytes In Vitro

Abstract

|
To determine which is better source of serum for maturation media in terms of promoting oocyte maturation in vitro and embryo development, immature oocytes obtained in excess of the number required by 12 patients with polycystic ovarian syndrome
were
randomized and cultured in vitro in a maturation media supplemented with human fetal cord serum (group ¥°) or preovulatory maternal serum (group ¥±). After maturation in vitro, oocytes of both groups were inseminated with husband's sperm and
evaluated
for fertilization and cleavage in vitro. In group ¥°, immature oocytes demonstrated a 46.7% (7/15) cleavage rate, and six among seven embryos developed normally and reached morula or blastocyst stage. In group ¥±, immature oocytes demonstrated a
53.3%
(5/18) cleavage rate, and seven among eight embryos normally and reached morula or blastocyst stage. No significant differences were noted between any of these parameters.
